Abstract

[Objective] To evaluate the effect of magnetic nanoparticle containing 5-fluorouracil (5-FU) targeting to treat hepatoma, cholangiocarcinoma and pancreatic carcinoma based on internal magnetic field built inside the tumor; [Methods] Internal magnetic field was produced by inserted magnetic biliary stent wires into tumor. 144 nude mouse bearing tumor were divided into three groups including hepatoma group (40 modes), cholangiocarcinoma group (64 modes) and pancreatic cancer group (40 modes). One sub-group in tumor group was treated by both magnetic nanoparticles containing 5-FU and internal magnetic field. Control sub-groups only were treated by one or none of them. The tumor volume was measured every 3 days, totally six times from treatment started. Tumor tissues were observed by transmission electron microscope after observation periods. [Results] Three experimental sub-groups show significantly therapeutic efficacy, Repeated Measures Analysis of Variance. Moreover, more apoptosis of tumor cells could be easily detected in those sub-groups. [Conclusion] Magnetic particles containing 5- FU combining with internal magnetic field could effectively treat hepatoma, cholangiocarcinoma and pancreatic carcinoma in targeting-site.
