| Abstract |
[Objective] When traditional Chinese medicine is diminished to nanosize, its bioavailability will be improved. Panax notoginseng root is highly prized for its supposed hemorheological properties and widely used in China. The objective of the present study was to prepare nano Panax notoginseng particles and study the physicochemical properties of Panax notoginseng. [Methods] Nano Panax notoginseng particles were successfully prepared by high speed centrifugal sheering pulverizer. The influence of processing parameters on the size of Panax notoginseng particle and the different properties of nano particles and raw powder were characterized using particle size analysis, scanning electron microscope (SEM) and transmission electron microscope (TEM). The content of effective ingredient Panax saponin Rb1 of Panax notoginseng before and after being ultrafinely ground were determined by reversed high performance liquid chromatography (HPLC). In addition, taking haemorrhage and coagulation time as index, the influences of different Panax notoginseng particle sizes on mice haemorrhage and coagulation system were studied. [Results] General Panax notoginseng powder could be ultrafinely ground to about 130 nm within 50 min, the cellular tissues of Panax notoginseng were destroyed, the active ingredients could dissolve out directly and fully, and the extractive ratio of Panax saponin Rb1 was increased 58.6% by high speed centrifugal sheering processing. The difference of haemorrhage and coagulation time of nano Panax notoginseng particles was statistically significant (P<0.05) compared with raw powder. [Conclusions] It is possible to prepare nanoparticles using high speed centrifugal sheering technology, and it is valuable to combine nanotechnology and TCM to improve the bioavailability and rapid releasing. |
